As someone who interviews and hires people for jobs, please read the ad and make sure you tailor your resume to job ad.
Don't send in a generic resume because that just wastes your time and my time.
If you don't have the skills, then see what is involved in learning the skills needed for that job.
Use the time between sending in the resume and the interview to learn any skill necessary.
I once got hired for a job because I was the only person who they interviewed who had bothered learning what structural equation modelling was, and was able to talk intelligently about it in the interview...
I was very upfront that I had not done any, but had engaged in similar matrix programming.
Actually by the time of the interview, I had read enough about it so that I knew more about it than the interviewer.
I also showed them some samples of my programming to make it clear that I can learn any syntax needed to run the software.
Be strategic in your job quest... get the skills you need to get the job you want.
